ANTIBODIES TO CHD PROTEINS

The CHD family of proteins appears to be primarily involved in the regulation of developmental gene expression and heterochromatin formation. There are nine members of the CHD family of proteins (CHD1-9). The CHD family members possess three common structural and functional domains: a chromodomain (chromatin organization modifier), an SNF2-like helicase/ATPase domain, and a C-terminal DNA-binding domain. The chromodomain functions to recognize and bind nucleosomes while the SNF2-like helicase/ATP domain provides the energy required for histone displacement and nucleosomal sliding for remodeling. The DNA-binding domain is more divergent among the CHD family members and may partially function to promote sequence specific interactions with DNA. Despite their structural similarity, the functions of the CHD family members are diverse. Functional diversity is likely imparted by the divergence in the DNA binding domain, as well as differences in binding partners and expression patterns of the CHD family members. Continued study of the functional contribution of each CHD family member to chromatin remodeling will further our understanding of how appropriate gene activation and repression is achieved in the developing organism.

Detection of Human and Mouse CHD4/Mi2 beta by Western Blot (h&m) and Immunoprecipitation (h).
  Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 mcg) and mouse NIH3T3 (M; 50 mcg) cells.  Antibodies: Affinity purified rabbit anti-CHD4/Mi2 beta antibody A301-082A used for WB at 0.04 mcg/ml (A) and 0.1 mcg/ml (B) and used for IP at 3 mcg/mg lysate.  CHD4/Mi2 beta was also immunoprecipitated by rabbit anti-CHD4/Mi2 beta antibodies A301-081A and A301-083A, which recognize other epitopes.  For blotting immunoprecipitated CHD4/Mi2 beta, ReliaBLOT® Reagents and Procedures (Cat. No. WB120) were used.  Detection: Chemiluminescence with exposure times of 10 seconds (A and B).

No. WB120) were used.  Detection: Chemiluminescence with exposure times of 30 seconds (A and B).


Detection of Human CHD1 by Western Blot and Immunoprecipitation.  Samples: Whole cell lysate from HeLa (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) and 293T (T; 50 mcg) cells.  Antibodies: Affinity purified rabbit anti-CHD1 antibody A301-218A used for WB at 0.04 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate.  CHD1 was also immunoprecipitated by rabbit anti-CHD1 antibody A301-217A, which recognizes an upstream epitope.  For blotting immunoprecipitated CHD1, ReliaBLOT® Reagents and Procedures (Cat. No. WB120) were used.  Detection: Chemiluminescence with exposure times of 3 seconds (A) and 10 seconds (B).
